Dr. William Masters is a renowned fertility doctor who wants to study the sexual lives of Americans. The year is 1956, so when his colleagues hear about his study, they warn him that he'll be labeled a pervert. He's confused about the habits and rituals in the bedroom in his studies as well as his relationship with his wife. They've been trying to get pregnant for years. Masters realizes that he can't explain the mysteries of the female in the equation without a woman's perspective. He hires Virginia Johnson. She's been married twice and used to sing in a nightclub before becoming a secretary. The two work to decipher the intricacies of human sexuality together while dancing around the developing feelings they have for each other.
